Caractérisation structurale multi-échelle de l’os à l’état précoce de l’ostéosarcome et modélisation de la morphologie tumorale à l’état avancé : observations de l’activation tumorale dans l’ostéogénèse

Abstract : Osteosarcoma is the main bone primary tumor, mainly afflicting a young demographic. Though the illness is remains rare, consequences are dire, with still to this day 2cm margins of excision and a long-term death rate after treatment of the order of 30%. Osteosarcoma is characterized by the creation of a mesh-like bone structure which grows alongside tumor cells progression. This cancer causes a degradation and production of bone tissue through a mechanism of bone remodelling hijacking called "vicious circle". In this thesis, we approach the study of osteosarcoma through physics analysis methods, the goal being to explore new opportunities to study this illness. We studied the bone structure at different development times, different scales and with different technical approaches. We participated in the uncovering with polarized light microscopy of the Bouligand structure of the bone construction interface. We used the nanoscopic structure of mineral and biological phases, revealed through X-ray diffraction, to put forward a model of early stage osteosarcoma activity. And we established a late stage osteosarcoma tomography acquisitions analysis protocol, which enables objective comparison with the results from the cellular automaton developped for the search of the osteosarcoma propagation laws.
